叶酸与鲱鱼精DNA作用机制的光谱研究

     

摘要

The interaction between folic acid(FA)and herring sperm DNA(DNA)was investigated by fluorescence and UV-vis spec-troscopy. The effects of DNA thermal denaturation,ionic strength,anion quencher KI and metal ion to the FA and DNA system were studied. The interaction models of FA and DNA were analyzed. The binding constants(K),binding sites(n)and thermodynamic pa-rameters at different temperatures were obtained. The results showed that the intrinsic fluorescence of FA could be quenched by DNA through static quenching mode. And the major binding modes between FA and DNA were groove binding and electrostatic binding.%采用荧光光谱和吸收光谱法研究了叶酸(FA)与鲱鱼精DNA(DNA)的相互作用。研究了DNA热变性、离子强度、阴离子猝灭剂KI及金属离子对FA-DNA体系的影响,分析了FA与DNA的相互作用模式,获得了不同温度下的结合常数、结合位点数及热力学参数。结果表明,DNA对FA的荧光有明显的猝灭作用,作用过程为静态猝灭,FA通过沟槽模式和静电作用力与DNA结合。
