PEG-无机盐双水相体系萃取水榆花楸叶中总黄酮

         

摘要

Flavonoids from sorbusalnifolialeaves had some pharmaceutic function.Aqueous two-phase extraction was an attractive method for flavonoids purification from the Sorbusalnifolia leaves.In this paper,two phase properties of aqueous two-phase system composed of various inorganic salt and different molecular weight PEG were studied.Besides,the effects of mass fractions of PEG and Na2SO4,temperature,pH and the amount of KCl on distribution behaviors of PEG1000-Na2SO4aqueous two-phase system was investigated when it was used to separate and purified the flavonoids from rowan,and test conditions were also optimized by orthogo-nal experiment.The results showed that optimum conditions were:mass fractions of PEG and Na2SO4were 15 and 30%,respective-ly,pH was 9,extraction temperature was 25℃ and the amount of KCl was 3%.The flavonoids extraction rate from rowan could reach 97.01%.Therefore,aqueous two-phase extraction was a high efficient method for flavonoids separation and purification from sorbusalnifolia leaves.%水榆花楸叶中所含的黄酮类化合物具有一定的药用价值,利用双水相技术萃取水榆花楸叶中的黄酮是一种有效的分离纯化方法.本文考察了不同种类的无机盐与不同相对分子量的PEG组成的双水相体系的两相性质,研究了PEG1000-Na2SO4双水相体系中分离纯化水榆花楸黄酮时PEG的质量分数、Na2SO4的质量分数、温度、pH、KCl添加量这5个因素对分配行为的影响,并通过正交实验优化了试验条件.结果表明最佳双水相萃取条件为:Na2SO4的质量分数为15%,PEG1000质量分数为30%,pH为9,提取温度为25℃,KCl的添加量为3%,此时原料中黄酮的萃取率可高达97.01%.因此,该体系是一种高效分离纯化水榆花楸叶中黄酮的方法.
