With green biodegradable polymer chitosan as substrate,genipin as the crosslinking agent,through observing whether the proper chitosan hydrogels could be made under normal pressure on different conditions,such as crosslinking temperature,crosslinking ratio and crosslinking reaction time,the optimal conditions were found out.Experiment showed that the hard hydrogels could be made only on the conditions of 37C of temperature,0.25% or 0.5% of crosslinking ratios,and 3h of crosslinking time.Chitosan porous scaffold was prepared by supercritical carbon dioxide foaming method.Infrared spectrum analysis and scanning electron microscopy results showed that when the crosslinking ratio was 0.25%,with the increase of pressure,the degree of crosslinking of chitosan and genipin also increased.When the pressure was on the 100bar,the best porous chitosan scaffold for tissue engineering could be obtained.Its pores are more uniform and it has appropriate strength.The porosity and water absorption of the scaffold are 92% and 571% respectively.%以绿色生物可降解高分子材料壳聚糖为基材,京尼平为交联剂,在常压下,通过考察制备过程中不同的交联温度、交联比和交联时间来确定能否形成合适的水凝胶,实验表明:温度为37℃,交联比为0.25%或0.5%,交联时间达3h时,可得到结实的水凝胶.采用超临界二氧化碳发泡法制备壳聚糖多孔支架材料,红外光谱分析及扫描电镜结果表明交联比为0.25%,壳聚糖与京尼平的交联程度随压力增大而提高,当压力为100bar时,可获得孔隙较为均匀且具有适宜强度的多孔壳聚糖组织工程支架,该支架的孔隙率和吸水率分别为92%和571%.
