Objective:To establish a method for the simultaneous determination of cucurbitacin B (CuB) and cucurbitacin E(CuE) in cucurbitacin tablets by reversed-phase high performance liquid chromatography.Methods:The Diamasil TM C18 column (250 × 4.6 mm,5 μm) was used for separated.The mobile phase consisted of 0.1% aqueous formic acid-acetonitrile,the flow rate was 1.0mL ·min-1.The detection wavelength was 254 nm and the column temperature was 35℃.Results:The cucurbitacin B and cucurbitacin E were completely separated in the cucurbitacin tablet within 40 min.The linearity of the CuB and CuE was linear with the mass concentration of the cucurbitacin and the peak area of the chromatogram.The linear equations of CuB and CuE were Y=8998X-18.38(r=0.9995) and Y=10960X-73.14 (r=0.9999) respectively.The average recoveries of CuB and CuE were 97.34% (RSD=2.71%),96.68% (RSD =2.33%).Conclusion:The method is simple and sensitive,and the results are accurate and reliable.The sample recovery rate is high,This method is suitable for the determination of cucurbitacin B and cucurbitacin E in tablets.%目的:建立反相高效液相紫外法同时测定葫芦素片剂中葫芦素B、葫芦素E的含量测定方法.方法:采用Diamasil TM C18柱(250×4.6mm,5μm),以0.1%甲酸水-乙腈为流动相,流速为1.0mL· min-1,检测波长为254 nm,柱温为35℃.结果:在40 min内,葫芦素片中葫芦素B(CuB)和葫芦素E(CuE)完全分离,进样的质量浓度与色谱峰面积呈现良好的线性关系,CuB及CuE的线性方程分别为:Y=8998X-18.38(r=0.9995),Y=10960X-73.14(r=0.9999),CuB及CuE的平均加样回收率分别为97.34%(RSD=2.71%)、96.68%(RSD=2.33%).结论:该方法操作简便灵敏、结果准确可靠,样品回收率高,适用于片剂中葫芦素B和葫芦素E的含量测定.
