bFGF2抑制BMP9诱导小鼠间充质干细胞C3H10T1/2向成骨细胞分化

         

摘要

Objective Observe the effect of FGF2 in BMP9-induced osteogenic differentiation of C3H10T1/2 mesenchymal stem cells,and investigate its mechanism.Methods 3H10T1/2 cells were infected by adenovirus recombined with FGF2 and BMP9,then the early osteogenic maker alkaline phosphatase(ALP) activity was detected by quantitative and staining assay,later osteogenic maker calcium deposition was determined by Alizarin Red S staining,pl2SBEluc activity was detected by quantitative,Smad1/5/8,Runx2 and OCN were determined by Western blot.Results BMP9-induced early osteogenic marker ALP,and late osteogenic markers matrix mineralization and osteocalcin (OCN) was inhibited by FGF2,BMP-Smad1/5/8 pathway and Runx2 were inhibited by FGF2.Conclusions FGF2 inhibits bone morphogenetic protein9-induced osteogenic differentiation of C3H10T1/2 mesenchymal stem cells.%目的 观察碱性成纤维细胞生长因子(FGF2)对于骨形态发生蛋白9(BMP9)诱导的间充质干细胞C3H10T1/2成骨分化的影响,并探讨其机制.方法 用BMP9和FGF2重组腺病毒感染C3H10T1/2细胞,测定成骨早期标志物碱性磷酸酶(ALP)活性变化,茜素红S染色观察成骨晚期标志物钙盐沉积,pl2SBE-luc荧光素酶报告基因活性检测,Western blot检测经典信号通路smadl/5/8和成骨关键转录因子RUNX2的变化,以及晚期成骨标志物骨钙素(OCN)的表达.结果 FGF2抑制了BMP9诱导的早期成骨指标ALP和晚期成骨指标钙盐沉积和骨钙素的表达,抑制了经典的BMPs-Smad信号通路,抑制了成骨关键的转录因子Runx2的表达.结论 FGF2可抑制BMP9诱导的间充质干细胞C3H10T1/2的成骨分化.
