蛇床子素长循环脂质体的制备与表征

         

摘要

Using ethanol injection method to prepare osthole liposomes(Ost-lips),then,osthole-loaded long-circulating liposomes(Ost-LCL)were formulated by adding PSPE-PEG 2000,50℃and incubate for 1 h,and the physicochemical properties of the two kinds of liposomes were compared and evaluated.The encapsulation efficiency of osthole in Ost-Lips and Ost-LCL was measured to be(83.09 ±0.56)% and (86.27 ±0.51)%,respectively.The size distribution were(99.6 ±3.1)and(124.3 ±3.8)nm,respec-tively.The mean value of Zeta potential were(-7.58 ±0.57)mV and(-24.21 ±0.40)mV,respec-tively.Ost-Lips was completely released at 24 h,with a cumulative release of 84.23%,and a complete re-lease of Ost-LCL at 48 h,with a cumulative release of 80.21%.Ost-LCL showed better stability and had better sustained-release effect in vitro release.%采用乙醇注入法制备蛇床子素脂质体(Ost-Lips),加入DSPE-PEG 2000,50 ℃孵育1 h,制得蛇床子长循环素脂质体(Ost-LCL),并对比评价两种脂质体的理化性质.结果表明,Ost-Lips和Ost-LCL的包封率分别为(83.09 ±0.56)%、(86.27 ±0.51)%;平均粒径(99.6 ±3.1),(124.3 ±3.8)nm;平均电位(-7.58 ±0.57),(-24.21 ± 0.40)mV;Ost-Lips在24 h基本完全释放,累计释放84.23%,Ost-LCL在48 h基本释放完全,累计释放80.21%.Ost-LCL表现出更好的稳定性,在体外释放中有更好的缓释效果.
