野菊花总黄酮对酒精致急性肝损伤小鼠的保护作用

     

摘要

目的 观察野菊花总黄酮(total flavonoids Chrysanthemum indicum,TFC)对酒精致小鼠急性肝损伤的保护作用及部分机制.方法 用TFC(125、250、500 mg·kg-1)连续给小鼠灌胃7 d,末次给药后1 h,一次性灌胃酒精(50%,6 g·kg-1)致小鼠急性肝损伤模型,测定不同剂量的TFC对急性肝损伤模型小鼠血清丙氨酸转氨酶(ALT)、天门冬氨酸转氨酶(AST)活性、甘油三酯(TG)含量和肝匀浆中丙二醛(MDA)含量及超氧化物歧化酶(SOD)、谷胱甘肽(GSH)、谷光甘肽过氧化物酶(GSH-PX)活性的影响;放射免疫分析法测定肝匀浆中肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)的表达;同时对肝组织进行病理学检查.结果 TFC能显著降低酒精致小鼠急性肝损伤血清ALT、AST、TG值,降低肝匀浆中MDA含量,增强SOD、GSH、GSH-PX活性,使TNF-α和IL-1β表达降低,并减轻酒精对肝组织的病理损伤.结论 TFC对酒精致小鼠急性肝损伤具有一定的保护作用,其机制可能与其清除自由基、抑制脂质过氧化、抑制TNF-α和IL-1β的表达有关.%Aim To study the protective effec:ts and mechanisms of total flavonoids Chrysanthemum indicum( TFC )on alcohol-induced acute liver injury in mice. Methods The animal model of acute injury was established by one dose of alcohol( 50% ,6 mg ·kg-l ,ig ).The serum transaminases( ALT.AST )activity and TG contents, MDA contents and SOD . GSH,GSH-PX activities in hepatic tissue were detected. The expression of TNF-α,1L-1β in different groups were also determined by radioimmunoassay. Results TFC could significantly decrease the activities of ALT and AST and TG content in serum, markedly decrease MDA and increase SOD, GSH, GSH-PX activities in hepatic tissue. The expressions of TNF-α and 1L-1β were also inhibited by TFC. Pathohistological observation indicated the liver lesion was markedly alleviated hy TFC administration. Conclusion These results suggest that TFC inhibits ALT and AST activities.decreases TG and MDA and up-regulates SOD, GSH , GSH-PX activities . which might be molecular mechanisms of its protective effects on alcohol-induced acute liver injury in mice.
