HPLC法测定络石藤中络石苷元的含量

摘要

Aim To establish a method for determination the content of trachelogenin in Trachelospermum Jasminoides (Lindl. ) Lem.Methods The symmetry C18 column(3. 9 mm x 150 mm,5 μm) was used . the mobile phase consisting of methyl-alcohol-water( 45 ∶55) . The flow rate was 0. 8 ml · min -1 ,the detection wavelength set at 280 nm and contents calculated with external reference method.Results This method showed good linear relation in the range of 5. 07～ 81. 12 mg · L-1 for trachelogenin ( r = 0. 999 7). The average recovery was 99. 76％ with RSD of 0. 83％ . Conclusion The method is simple , quick, accurate and suitable for the quality control of Trachelospermum Jasminoide.s ( Lindl. ) Lem.%目的 建立测定中药络石藤中络石苷元含量的高效液相色谱方法.方法 甲醇-水(7∶ 3)提取药材,经酸水解、碱水解后调pH值为 6.0沉淀用甲醇溶解,高效液相色谱法测定,Symmetry C18(3.9 mm×150 mm,5 μm),流动相甲醇-水(45∶ 55),流速0.8 ml·min-1,检测波长280 nm,采用外标法峰面积计算含量.结果 络石苷元溶液在5.07～81.12 mg·L-1时与其吸收值呈良好线性关系(r=0.999 7),平均加样回收率为99.76%,RSD=0.83%.结论 该方法操作简便、快捷,结果准确、可靠,适用于络石藤中络石苷元的含量测定.
