Objective To explore whether lupeol could prevent GVHD through DC suppression.Methods DCs were prepared from mouse BM progenitor in the presence of GM-CSF/IL-4 and expanded in vitro.The effects were observed of lupeol on DCs.We performed flow cytometry to study the phenotypic changes of DCs,conducted mixed lymphocyte reaction(MLR)to assay allo-stimulatory capacity of DCs.Also performed ELISA to study the secretions of TNF- αand IL-012 in the cellular supernatant in DCs.Results Lupeol inhibited the up-regulation of surface markers CD80,CD86,CD40 as well as MHC-II in DCs.Lupeol reduced the capacity of DCs to induce allogeneic lymphocytes proliferation in MLR.Lupeol also substantially affected the capacity of DCs to produce TNF-α and IL-12.Conclusion DC suppression could be one of the mechanisms that administration of lupeol could prevent GVHD.%目的 体外研究羽扇豆醇在移植物抗宿主病中的作用.方法 培养骨髓来源的树突状细胞(DC),在加入羽扇豆醇后,流式检测细胞表面分子、ELISA法检测培养上清细胞因子,混合淋巴细胞反应(MLR)检测异基因淋巴细胞增殖.结果 羽扇豆醇作用后,DC细胞表面分子下调;TNF-α和IL-12分泌减少;异基因淋巴细胞增殖受到抑制,呈现出明显的剂量依赖性.结论 羽扇豆醇抑制了DC的成熟和活化,此将在GVHD中发挥作用.
