低分子肝素及HB-EGF对孕早期滋养细胞功能的影响

         

摘要

Objective To investigate the efficacy of modulation of low molecular weight heparin and heparin-binding epidermal growth factor on functions of human first trimester trophoblast. Methods Vitro trimester trophoblast cells were chosen as research objects, which were incubated with different concentrations of heparin and different drugs. And the trophoblast proliferation,invasion and HCG secretion were compared. Results When the concentrations of low molecular weight heparin were 0. 25 U·mL-1 and 2. 5 U·mL-1 , it promoted the first trimester trophoblast cell proliferation and invasion(P<0. 05),while it rose to 25 U·mL-1 and 250 U·mL-1, low molecular weight heparin significantly reduced trophoblast invasion and proliferation(P<0. 05). The combined group was stronger than the other two groups in promoting trophoblast cell invasion and proliferation(P<0. 05). Low molecular weight heparin inhibited trophoblast cells from secreting HCG,while the HB-EGF group and the joint group promoted HCG secretion(P<0. 05). ConclusionsLow molecular weight heparin plays an important role in the first trimester trophoblast invasion,proliferation and secretion,and HB-EGF promotes the role of low molecular weight heparin.%目的:探讨低分子肝素及肝素结合型表皮生长因子( HB-EGF)对孕早期滋养细胞功能的影响。方法选取体外培养的孕早期绒毛滋养细胞进行研究,采用不同浓度肝素及不同用药搭配进行培养,比较其滋养细胞增殖、侵袭和HCG分泌能力。结果低分子肝素浓度在0.25 U·mL-1和2.5 U·mL-1时则对孕早期滋养细胞增殖和侵袭能力起促进作用(P<0.05),而在25 U·mL-1及250 U·mL-1时,低分子肝素明显降低滋养细胞侵袭及增殖能力(P<0.05)。联合组对增殖能力和侵袭能力的促进强于另外两组(P<0.05)。低分子肝素组对滋养细胞HCG分泌起抑制作用,而HB-EGF及联合组对HCG分泌具有促进作用(P<0.05)。结论低分子肝素对孕早期滋养细胞的侵袭、增殖以及HCG分泌具有重要作用,而HB-EGF对低分子肝素发挥功能起促进作用。
