叶黄素对博来霉素所致的小鼠肺纤维化治疗作用及其对小鼠氧化应激水平影响的研究

         

摘要

Objective To explore the effects of lutein on bleomycin-induced lung fibrosis in mice,providing certain reference for the application of this compound.Methods Eighty KM mice were randomized into normal control group (normal saline),positive group (prednisone acetate,0.45 mg · kg-1 · d-1),high dose of lutein group (200 mg · kg 1 · d-1),middle dose of lutein group (100 mg · kg-1 · d-1) and low dose of lutein group (50 mg · kg-1 · d-1).After 21 days,the blood and lung were obtained.The lung coefficient,levels of MDA,GSH-PX,SOD and T-Aoc were detected.The HE staining was performed and observed in mice.The method of Szapiel was used to evaluate alveolar inflammation and fibrosis levels in mice.Results The lung coefficient,MDA,alveolar inflammation and fibrosis scores in normal control,positive group,high dose and middle dose group were significantly lower than the model group,while the levels of GSH-PX,SOD and T-Aoc were obviously higher than the model group(Flung index =4.892,FMDA =4.761,Finflammation scores =4.692,Ffibrosi scores =4.502;FGSH-PX =4.437;FSOD =4.381;FT-Aoc =4.821,all P < 0.05).Pathological section indicated that the pulmonary alveoli and tissues in the above groups were normal,while those in the model group had significant pathological changes.All the above indicators in low dose group were consistent with the model group.Conclusions Lutein has good treatment effect on bleomycin-induced pulmonary fibrosis in mice and is worthy of further study.%目的 探讨叶黄素对博来霉素所致的小鼠肺纤维化的治疗作用及其对小鼠氧化应激影响,为该化合物的应用提供一定的借鉴.方法 昆明小鼠80只,随机分为正常组(等量生理盐水)、阳性药组(醋酸泼尼松,剂量为0.45 mg·kg-1·d-1),叶黄素高、中及低剂量组每天灌胃给予叶黄素,剂量分别为200、100及50 mg·kg-1·d-1.给药21 d后,取血及肺,计算肺系数;检测血清及肺匀浆中丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-PX)、超氧化物歧化酶(SOD)及总抗氧化能力(T-Aoc)含量;利用苏木精-伊红染色法,观察小鼠肺病理切片并利用Szapiel法对各组小鼠的肺泡炎及纤维化程度进行评价.结果 正常组、阳性药组、高剂量及中剂量组小鼠肺系数、MDA含量、肺泡炎及纤维化评分均明显低于模型组,而GSH-PX、SOD及T-Aoc水平明显高于模型组(F肺系数=4.892,FMDA =4.761,F肺泡炎评分=4.692,F纤维化评分=4.502;FGSH-PX=4.437;FSOD=4.381;FT-Aoc=4.821,均P<0.05);病理切片显示,上述各组小鼠肺泡及组织基本正常,而模型组则均出现明显病变;低剂量组上述指标均与模型组基本一致.结论 叶黄素对博来霉素所致肺纤维化小鼠具有较好的治疗作用,对于该化合物的应用具有一定的借鉴意义,值得进行深入研究.
