Objective To study the effect of the changing expression of Grim-19 on the telomerase activity in HeLa cell which was transfected from recombinant plasmid. Methods After the plasmid p3 x flag-myc-cmv-24-Grim-19 ( Grim-19 ) was transfected into the HeLa cells, Western blot were used to test the expression levels of Grim-19, htert,P16 and P21 after 48 h. The senescence and telomerase activity of HeLa cell were also detected. Results The low expression of flag and htert and overexpression of PI 6 and P21 can be obvious after the transfection, and the increasing cellular senescence and weakening proliferation and reducing telomerase activity were also found to be-true. Conclusion Grim-19 can regulate the telomerase activity and cell senescense in the cervical cancer cell line.%目的 重组质粒转染HeLa细胞以探讨Grim-19表达水平的变化对HeLa细胞中端粒酶活性的影响.方法 将重组质粒p3×flag-myc-cmv-24-Grim-19(简写Grim-19)通过脂质体转染至HeLa细胞,48 h后通过Western blot检测Grim-19、htert、P16、P21的表达情况,同时检测HeLa细胞衰老和端粒酶活性.结果 转染后可见flag的表达及htert的表达降低及P16、P21的表达升高,细胞衰老增加,增殖减弱及端粒酶活性的降低.结论 在子宫颈癌细胞株HeLa中Grim-19具有调控端粒酶活性和细胞衰老的作用.
