To explore the associations of human leukocyte antigen ( HLA )-DQAl region allele with the outcomes of hepatitis B virus ( HBV ) infection in Uygur population of Xinjiang. Methods Polymerase chain reaction-sequence specific primers ( PCR-SSP ) was used to genotype HLA-DQA1 gene. 70 cases chronic hepatitis B patients,46 cases chronic asymptomatic HBV carriers,42 cases spontaneous recovery after HBV infection and 80 cases healthy controls were recruited. The frequency distributions of genotype among four groups were analyzed. Results The frequency of HLA-DQA1 * 0102 in spontaneous recovery group and healthy controls group was significantly higher than those in chronic hepatitis B patients and chronic asymptomatic HBV carriers ( P < 0. 05 ). The frequency of HLA-DQA1 * 0501 in chronic asymptomatic HBV carriers was higher than those in healthy controls group ( P <0. 05 ). In addition, the frequency of HLA-DQA1 * 0501 in chronic hepatitis B patients was markedly higher than those in spontaneous recovery group and healthy controls group ( P < 0. 05 ). The frequencies of other DQA1 alleles were not significantly different among the groups. Conclusions The subjects with HLA-DQA1 * 0501 allele have an increased risk to chronic hepatitis B infection compared with other subjects without this allele, while HLA-DQA1 * 0102 carriers may reduce the risk of chronic hepatitis B occur in Uygur population of Xinjiang.%目的 探讨新疆维吾尔族人类白细胞抗原(HLA)-DQA1区等位基因多态性与乙型肝炎病毒(HBV)感染临床转归的相关性.方法 应用序列特异性引物-聚合酶链反应技术(PCR-SSP)对70 例慢性乙型肝炎患者,46 例HBV携带者,42 例HBV感染后自然恢复者,80例健康献血员进行HLA-DQA1基因分型,并比较DQA1多态性的分布频率及其差异.结果 HBV感染后自然恢复组与健康对照组HLA-DQA1*0102分布频率高于慢性乙型肝炎组和HBV携带者组,差异均有统计学意义(P<0.05);HBV携带者组HLA-DQA1*0501分布频率高于健康对照组,差异有统计学意义(P<0.05);慢性乙型肝炎组HLA-DQA1*0501分布频率高于HBV感染后自然恢复组与健康对照组,差异有统计学意义(P<0.05);其他各等位基因频率在各组间差异无统计学意义.结论 新疆维吾尔族人群中携带HLA-DQA1*0501等位基因者感染HBV后可能会增加慢性乙肝发生的风险,而携带HLA-DQA1*0102者可能会降低慢性乙肝发生的风险.
