Objective To investigate the change of SnoN expression in hippocampus of SD rats after global cerebral ischemia/reperfusion,and to elucidate the impiication of SnoN in ischemic neuronal apoptosis. Methods Thirty adult SD male rats were randomly divided into 2 groups: sham group(l0) and ischemia/reperfusion (I/R) group ( 20) . Global cerebral ischemia/reperfusion model was produced by Pulsinelli 4-VO method. Immunohistochemical staining was performed to detect neuron and SnoN expression. SnoN and TGF-β protein expression was detected by Western blot. Results Immunohistochemistry and Western blot revealed that the expression of SnoN was significantly increased(P<0. 05)with extensive neuronal loss on the 1st and 3rd day after global cerebral ischemia/reperfusion. The TGF-β pathway was activated. Conclusion The overexpression of SnoN in rat hippocampus after global cerebral ischemia might be involved in ischemic neuronal loss through activation of TGF-β pathway.%目的 通过观察全脑缺血大鼠海马组织中转录共轭因子SnoN表达水平的变化,探讨其在脑缺血中的作用.方法 30只雄性SD大鼠随机分成手术组和假手术组,假手术组、手术1、3 d组各10只.手术组采用Pulsinelli四血管结扎法建立大鼠全脑缺血再灌注模型,假手术组仅暴露第一椎间孔及颈动脉.免疫组织化学法检测海马组织神经元及SnoN的表达;Western blot检测SnoN及TGF-β蛋白水平的变化.结果全脑缺血再灌注后1、3 d海马CA1区部分神经元缺失,与假手术组相比,手术组术后1、3 d海马区SnoN、TGF-β表达量明显升高(P<0.05).结论 大鼠全脑缺血后海马组织SnoN表达增高,可能通过激活TGF-β通路参与神经元缺血性凋亡过程.
