溶血磷脂酸对脊髓星形胶质细胞增殖的影响

         

摘要

Objective To observe the effect of lysophosphatidic acid (LPA ) on spinal astrocytic proliferation in vitro. Methods Primary cultured rat spinal astrocytes were randomly divided into two groups : control group (treated by DMEM/F12 containing 10% FBS)and LPA group(treated by DMEM /F12 containing 10% FBS and 10 μmol/L LPA). At different time points (0 ,6 ,12 ,24 and 48 h) ,the number of cells was counted using a hemocytometer ,the expression changes of proliferating cell nuclear antigen (PCNA)were evaluated by Western blot ,and the cell cycle was analyzed by using flow cytometry (FCM ). Results The purity of cultured spinal cord astrocytes was up to 98% as determined by immunofluorescent staining. There was no statistically significant difference in the cell proliferative number ,PCNA expression and cell cycle distribution between control group and LPA group at all time points (all P>0. 05). Conclusion LPA shows no effect on proliferation and cell cycle progression of spinal astrocytes in vitro.%目的 察溶血磷脂酸(LPA)对体外培养脊髓星形胶质细胞增殖的影响.方法 采用原代培养的大鼠脊髓星形胶质细胞,实验分为对照组(正常培养液培养)和LPA干预组(含有10 μmol/L LPA的培养液培养);在不同时间点(0、6、12、24和48 h),计数各组的细胞数目,采用Western blot和流式细胞术检测各实验组增殖细胞核抗原(PCNA)表达量变化及细胞周期进展情况.结果 免疫荧光染色显示原代培养的脊髓星形胶质细胞纯度达98%;在各观察时间点,对照组和LPA干预组的星形胶质细胞的细胞增殖数目、PCNA蛋白表达量及细胞周期分布情况均无显著差异(均P>0.05).结论 LPA对体外培养脊髓星形胶质细胞的增殖活力和细胞周期进展并无显著影响.
