双氢青蒿素下调Notch1蛋白表达对人卵巢癌Skov3细胞抑制作用的研究

         

摘要

目的:研究双氢青蒿素(dihydroartemisinin ,DHA)对人卵巢癌Skov3细胞增殖及对Notch1 mRNA、蛋白表达的影响。方法体外培养人卵巢癌Skov3细胞,加入不同浓度的DHA(分别为0、5、10、20、40、80μmol/L),采用MTT比色法检测不同浓度DHA对人卵巢癌Skov3细胞增殖的影响;采用RT-PCR法和Western blot法检测不同浓度DHA处理24 h的人卵巢癌Skov3细胞内Notch1 mRNA和蛋白表达水平的变化。结果不同浓度的DHA处理不同时间(24、48、72 h)后,卵巢癌Skov3细胞的生长明显受到抑制(P<0.05);随着DHA浓度的增加,人卵巢癌Skov3细胞内Notch1 mRNA和蛋白的表达逐渐下降(均 P<0.05)。结论 DHA可能通过下调 Notch1的表达抑制人卵巢癌Skov3细胞的增殖。%Objective To investigate the effects of dihydroartemisin (DHA ) on the proliferation of human ovarian cancer cell line Skov3 and on the expression of Notch1 mRNA and protein.Methods Skov3 cells (a human ovarian cancer cell line ) were cultured and treated with different concentration of DHA (0 ,5 ,10 ,20 ,40 ,80 μmol/L).MTT assay was used to measure the effects of different concentrationss of DHA on the proliferation of Skov 3 cells.RT-PCR and Western blot analysis were used to examine the expression level of Notch1 mRNA and protein in Skov3 cells treated with different concentrations of DHA for 24 h.Results The proliferation of skov3 cells was significantly inhibited by different concentrations of DHA treatment at each time point (24 ,48 ,72 h) (P<0.05).The expression level of Notch1 mRNA and protein in Skov3 cells was downregulated with the DHA concentration increasing.Conclusion DHA may suppress the proliferation of human ovarian cancer Skov3 cells by down-regulating the expression of Notch1.
