RP-HPLC法测定复方丹参降脂颗粒中牡荆素和金丝桃苷的含量

         

摘要

目的:建立复方丹参降脂颗粒中牡荆素和金丝桃苷的含量测定方法。方法:采用反相高效液相色谱法(RP-HPLC)进行测定,色谱柱为Eclipse XDB-C18色谱柱(250mm ×4.6mm,5μm),流动相为乙腈-甲醇-四氢呋喃-0.5%醋酸水溶液梯度洗脱,检测波长为360 nm ,柱温为32℃,流速为1.0 mL/min。结果:牡荆素和金丝桃苷分别在1300~2600ng(R2=0.9995)和1600~3200ng(R2=0.9997)范围内呈良好的线性关系,平均回收率( n=6)分别为99.0%和99.1%, RSD分别为2.32%和1.72%。结论:该方法准确可靠、重现性好,灵敏度高,可作为复方丹参降脂颗粒的质量控制标准。%Objective:To establish a quantitative method to determine the contents of Vitexin and Hyperoside in particles of compound salvia miltiorrhiza lipid .Methods:Reversed phase high-performance liquid chromatograph ( RP-HPLC) method was adopted.Vitexin and Hyperoside were performed in a Eclipse XDB -C18(250mm ×4.6mm, 5μm)column with a mobile phase of acetonitrile -methanol-tetrahydrofuran -0 .5% acetic acid water in a gradient elution mode , the detection wavelength was 360 nm, the column temperature was 32℃and the flow rate was 1.0 mL/min.Results:The linear ranges of Vitexin and Hyperoside were within 1 300~2 600ng(R2 =0.999 5) and 1 600~3 200ng(R2 =0.999 7).The average recoveries (n=6) were 99.0%and 99.1%, RSD 2.32%and 1.72%, respectively.Conclu-sion:This method is reliable , accurate , which has advantages of higher sensitivity and precision .It can be used for quality control of particles of compound salvia miltiorrhiza lipid .
