黄连与干姜配伍对黄连中5种生物碱的大鼠体内药动学影响

         

摘要

Objective:To investigate the plasma pharmacokinetics change of five main alkaloids ( berberine, palmatine, jatrorrhizine, coptisine and tableberberine) of coptidis in rats compared after the compatibility of coptidis and coptidis-ginger.Methods:Two groups were orally administered respectively by decoctions including decoction of coptidis and blend decoction of coptidis and ginger which were decocted separately.The contents of five alkaloids of coptidis in rats'plasma were detected and compared before and after the administration of the pharmacokinetic parameters.Results:The pharmacokinetic parameters obtained showed that C max and AUC0-t of five alkaloids of coptidis were increased .MRT0-t and t 1/2z were decreased and there was no obviously change in T max when coptidis is combined with ginger.Conclusion:Ginger can promote the absorption of berberine alkaloids ,and can also accelerate the elimination rat of each alkaloid rat. There are obvious effects on pharmacokinetic of five main alkaloids of coptidis in rats when coptidis is combined with gin-ger.%目的:考察黄连与干姜配伍后对黄连的5种主要生物碱(小檗碱、巴马汀、药根碱、黄连碱和表小檗碱)大鼠体内药动学的影响。方法:Wistar大鼠分别单次灌胃给予黄连单煎液和黄连干姜合并液后,运用HPLC法同时测定不同时间点的5种生物碱血药浓度,分别求算其药动学参数,比较配伍前后在大鼠体内药动学的变化。结果:黄连干姜合并液组与黄连单煎液组比较,黄连的5种主要生物碱的药时曲线下面积(AUC0-t)和峰浓度(Cmax)增加,半衰期(t1/2z)和平均滞留时间(MRT0-t)均降低,达峰时间( Tmax )无明显变化。结论:黄连与干姜配伍后能够促进黄连中生物碱在大鼠体内的吸收量和加速其消除速率,干姜能够显著影响黄连5种生物碱在大鼠体内的药动学。
