Objective:To investigate the influence of Buyang Huanwu decoction on apoptosis of microvascular endothelial cells induced by Aβin rats.Methods:Establish the damage models of microvascular endothelial cells induced by Aβ and different concentrations of Buyang Huanwu decoction (0.1,1,10 ug/ml) were given.Methylthiazolyl tetrazolium (MTF) assay was adopted to measure the viability of microvascular endothelial cells.Western blot method was used to determine the expression of bax and caspase-9 proteins.Results:The expression of bax and caspase-9 proteins of microvascular endothelial cells induced by Aβ were inhibited by Buyang Huanwu decoction (0.1,1,10 ug/ml) in a concentration-dependent manner.Conclusion:Buyang Huanwu decoction can inhibit the expression of bax and caspase 9 proteins and it has a protective effect on microvascular endothelial cells in rats,which may be an effective therapeutic agent for cardiovascular system diseases.%目的:研究补阳还五汤对Aβ诱导的大鼠微血管内皮细胞(PMVEC)凋亡的影响.方法:建立Aβ损伤大鼠微血管内皮细胞模型,不同浓度(0.1、1、10 μg/ml)补阳还五汤干预,MTT法检测大鼠微血管内皮细胞活力;western-blot法检测大鼠微血管内皮细胞中凋亡蛋白bax和caspase9表达水平.结果:补阳还五汤在浓度(0.1、1、10 μg/ml)能够抑制Aβ诱导的大鼠微血管内皮细胞bax和caspase9蛋白的表达并呈浓度依赖性.结论:补阳还五汤能够通过抑制凋亡蛋白bax和caspase9的表达而具有保护大鼠微血管内皮细胞的作用,可能成为是一种有效的抗脑血管系统疾病治疗剂.
