HPLC法测定槟榔中槟榔碱和槟榔次碱的含量

         

摘要

目的:建立高效液相测定槟榔中槟榔碱和槟榔次碱的含量测定方法.方法:采用HPLC法,色谱柱为Nucleosil SA阳离子交换色谱柱(4.6 mm×250 mm,5μm),流动相为甲醇-水-磷酸(60:40:0.3,氨试液调pH3.8),流速1.0 mL·min-1,检测波长为212 nm,柱温30℃.结果:槟榔碱和槟榔次碱与其他成分分离良好,槟榔碱在2.02~32.30μg·mL-1浓度范围内线性关系良好(R2=0.9995),平均回收率为97.2%,RSD为2.4%(n=9);槟榔次碱在0.85~13.55μg·mL-1浓度范围内线性关系良好(R2=0.9993),平均回收率为103.9%,RSD为2.6%(n=9).结论:所建方法专属性、准确,重复性好,可用于测定槟榔中槟榔碱和槟榔次碱的含量.%Objective:To establish an determination method for arecoline and arecaidine in Binglang( Semen Arecae) by HPLC. Methods:Nucleosil SA cation-exchange column(4. 6 mm × 250 mm, 5μm) was used with methanol-water-phosphoyic acid (60:40:0. 3, pH 3. 8) as the mobile phase at the flow rate of 1. 0 mL·min-1. The detection wave length was 212 nm and the column temperature was 30℃. Results: Arecoline and arecaidine were well isolated from other substances. For arecoline, there was a good linearity in the range of 2. 02~32. 30 μg·ml-1(R2 =0. 9995) and the average recovery rate was 97. 2% with RSD as 2. 4% (n=9). For arecaidine, there was a good linearity in the range of 0. 85~13. 55 μg·ml-1(R2 =0. 9993), and the average recovery rate was 103. 9% with RSD as 2. 6%(n=9 ) . Conclusion:The determination method is specific, accurate and repeatable, which can be used for determing areco-line and arecaidine in Semen Arecae.
