超声波辅助提取常春油麻藤总黄酮的工艺优化

         

摘要

以常春油麻藤为原料,在单因素实验的基础上,以总黄酮提取率为指标,采用正交实验L9(34)优化超声辅助乙醇回流提取总黄酮的最佳工艺条件。结果表明,最佳提取工艺参数为乙醇体积分数75%、料液比1∶30( g/mL)、超声提取温度80℃、超声提取时间55 min,在此条件下,测得常春油麻藤的藤、茎、叶、花中总黄酮的平均提取率分别为16.56%、17.05%、19.19%、20.31%。该方法操作简便、提取率高、重复性好,可用于常春油麻藤总黄酮的提取和含量的测定。%Using the extraction rate of total flavonoids from Mucuna sempervirens Hemsl as the target index, on the basis of the results of single-factor tests, we optimized the technological conditions for the extraction of total flavonoids by using ultrasonic-assisted ethanol reflux method through orthogonal test L9(34).The optimum extraction technological conditions were obtained as follows:the volume fraction of ethanol as extractant was 75%;the ratio of raw material to ethanol was 1∶30 ( g/mL); the ultra-sonic extraction temperature was 80℃;the ultrasonic extraction time was 55 min.Under these optimum conditions , the extraction rate of total flavonoids from the rattan, stem, leaf and flower of M.sempervirens was 16.56%, 17.05%, 19.19%and 20.31%, re-spectively.This method has the advantages of simple operation , high extraction rate and good repeatability.Therefore it can be used in the extraction of total flavonoids from M.sempervirens and the determination of total flavonoid content in this species of plant.
