目的:采用薄层色谱分离和生物活性检测相结合的薄层色谱-自显影技术,对石榴皮提取物清除自由基活性成分进行研究.方法:以三元溶剂二甲苯:乙酸乙酯:乙酸=3:5.5:1.5为展开剂对石榴皮提取物进行展开分离,以二苯代苦味肼自由基(DPPH)为显色剂,喷洒于经展开后的TLC进行显色拍照.结果:石榴皮提取物在传统显色剂下分离到6个化合物,利用DPPH自由基显色,表明其中4个化合物具有较强的清除作用,且活性成分主要为鞣质类化合物.结论:薄层色谱-自显影技术是一种快速分离鉴定天然产物中清除自由基活性成分的实验方法.%Objective:To investigate the free radical scavenging activity of pomegranate peel extracts by thin-layer chromatography ( TLC ) -bioautography.Methods: Pomegranate peel extracts were separated by compound developing solvents of xylene, ethyl acetate and acetic acid in ratio of 3∶5.5∶1.5 ) and sprayed with chromogenic agent of 1, l-diphenyl-2-picrylhydrazyl(DPPH) solution in ethanol for the photos.Results: Six different compound points appeared by classical staining reagents, while four free radical scavenger points were developed with DPPH solution in ethanol, suggesting that the 4 components, primarily compounds of tannis, were of strong scavenging activity.Conclusion:TLC- bioautography is a quick approach to separating and detecting free radical scavenging active components of natural products.
