Heterokaryosis is a common pheomenon in filamentous fungi especially in imperfect fungi and some ascomycetes. It is belived that heterokaryosis is one of the important source of fungal variation and the degeneration of crop disease-resistance. In 1960s', Dr. Ta Fuyu demonstrated that under natural conditions most of the bakanae disease of rice pathogen Fusarium moniliforme existed as a heterokaryons which consisted of 2-3 different type component nuclei. They differed from one another in certain cultureal morphological and physiological characters as well as pathogenicity. In our previous study, we attempted to distinguish the three different compoenent nuclear types from the dame strain of F.moniliforme heterokaryon by RAPD technique, the results showed that the DNA of the three type nuclei were highly homologous. In this paper, we report our recent study on DNA fingerpriting of three different type nuclei from F.moniliforme heterokaryon using 3 probes (33.6, M13 and pGTG4) combined with 2 restrition endonuclease (Hind III and Pst I) totally six combinations. The results of the experiments consists with those using RAPD analysis: the DNA of the three different component nuclear types of the F.moniliforme heterokaryon are highly homologous.
