It is generally thought that nuclear gene delivery is the most important process for gene expression. As a result, various non-viral vectors are currently being developed with this assumption in mind. However, our earlier study of quantitative analysis on the intracellular trafficking of pDNA revealed that a difference in the transfection activity between the viral and non-viral vectors principally resulted from intranuclear disposition of pDNA rather than its delivery to the nucleus. This difference is largely the result of the intranuclear disposition of pDNA rather than its delivery to the nucleus. In this study, we report on some direct evidence that demonstrates the importance of the release of intranuclear DNA from a vector on transfection activity. The data show that transfection activity is substantially enhanced by integrated system of multifunctional envelope-type nano device with biocleavable polyrotaxane for intranuclear DNA release.
