Most of conventional enzyme-responsive fluorescence probes are designed by masking afunctional group on fluorophores.1 However,the bulky fluorophore next to the enzymerecognition site decreases the rate of enzymatic reaction.Self-immolative spacers can enhancethe enzyme-responsiveness,2 but the process for the cleavage of a self-immolative spacerfollowed by the formation of byproducts which might affect biological system is inevitable.Inthis research,to enhance enzyme-responsiveness without producing byproduct,we havedeveloped a new type of enzyme-responsive probes CyME4-C and CyA4-C (Figure 1a).
