Intracellular environment remains highly crowded by biomacromolecules known as‘molecular crowding'.Molecular crowding affects important processes like replication,transcription and translation by altering the stability and structure of nucleic acids.1 Therefore,it is crucial to predict nucleic acid stability under the crowding condition to elucidate biologicalprocesses and develop technologies related to nucleic acids in cells.Nearest-neighbor (NN)model is the established concept,which can predict the stability by summing the NN pairs ofthe sequence of a duplex.2 However,the prediction in the crowding condition is still unexplored.
