摘要

In the previous research, RNA interference recombinant targeted KDR was successfully constructed, and effectively inhibited the expression of KDR gene in different kind of tumor cells, but exposed inefficiencies in the host cells transfected. In order to solve the problem efficiently, A TAT and KDR siRNA fusion gene lentiviral vector was constructed by recombinant technology and its effect on tumor cell membrane function and antitumor activity was observed through the experimental study in vitro. As a result, its anticancer effect is mainly manifested as that can effectively inhibit A549 cell KDR gene expression, cell proliferation and colony formation. In addition, it can also promote cell apoptosis and have strongly tumor targeting effect. So that, this lentiviral vector has significantly tumor target and antitumor activity.
