Isothermal nucleic acid amplification tests (NAATs) such as loop-mediated isothermal amplification (LAMP) show promise for use in point-of-care diagnostics in low-resource settings, because in principle these tests require simpler instrumentation and less power than PCR, and the enzymes are frequently more tolerant to inhibitors present in crude sample matrices. However, detection techniques lag behind PCR, particularly for multiplexed detection. In PCR, target-specific detection for multiple targets can be achieved using hydrolysis probes (e.g. Taqman probes), which are dependent upon the 5'-3' exonuclease activity of Taq DNA polymerase.
