Circulating cell-free DNA (cfDNA) is under investigation as an effective biomarker to monitor cancer oncogenesis, graft rejection, and infectious disease. Finding inspiration in recently-developed library preparation techniques in paleogenomics (ancient DNA and cfDNA are both damaged, single-stranded, and short), we utilized the ligation of single-stranded cfDNA to biotinylated probes (ss-ligation).
