Recently we developed a method where large antibody libraries are rendered infectious for eukaryotic cells, thereby directly linking the antibody genotype to the phenotype of individual cells. The power of the method derives from the fact that the potential agonists are selected from a large naive or preselected diversity system that contains about 10~8-10~(11) unique members. In the best case when autocrine screening systems are employed, one can analyze about two million phenotypic events per hour. Using these methods, a variety of agonist antibodies have been discovered that regulate cell fates, including those that are phenocopies of important cytokines as well as those that regulate the lineage specification and transdifferentiation of stem cells. The method only seems to be limited by the availability of selection systems.
