Almost all membrane and extracellular proteins are modified post-translationally by glycosylation. The glycan part of a protein becomes an integral part of the molecule, significantly affecting both structure and function. A recent comprehensive report endorsed by the US National Academies states that "glycans are directly involved in the pathophysiology of every major disease". Although often overlooked, glycosylation is important in IgG-class antibody effector function and has been shown to affect the pro-or anti-inflammatory nature of an IgG molecule. Changes in plasma protein or IgG glycome composition were found in many different diseases such as autoimmune diseases, diabetes and virtually all types of cancer.
