In many strategies to produce heterologous proteins, recovery of a pure protein of interest (POI) requires the researcher to lyse cells and purity the POI from the cellular milieu. To circumvent this requirement, we engineer a bacterial host for generalized secretion of a POI into the extracellular space, affording high yields of pure, folded protein. The type III secretion system (T3SS) of Salmonella enterica is comprised of a heterogeneous multi-protein structure that spans the periplasm, inner and outer membranes, and projects out from the cell, resembling a hypodermic needle.
