摘要

The assessment of protein stability is of key importance for drug development, drug discovery and basic research. Both the thermal and chemical stability of biologicals, such as antibodies, are routinely monitored to establish optimal conditions for large-scale production and long-term storage. In addition, shifts in the melting temperature (Tm) of a target protein upon binding to a small molecule ligand are used as indicators for binding events in primary screens during the early drug discovery process. Moreover, the precise analysis of protein unfolding and refolding mechanisms can yield important insights into the thermodynamic origins of protein folding, thereby helping to elucidate the molecular basis of degenerative diseases such as Alzheimer's, Parkinson's or diabetes.
