N-3-guanidinopropyl methacrylamide-N-2-hydroxypropyl methacrylamide copolymers (GPMA-HPMA) synthesized via radical copolymerization of N-3-aminopropyl methacrylamide (APMA) and N-2-hydroxypropyl methacrylamide (HPMA) followed by guanidination were designed to develop novel non-viral gene carriers with cell membrane-penetrating properties. Synthesized GPMA-HPMA copolymers complex with pDNA completely at charge ratios no more than 1, and the positively charged complexes have diameters about 200 nm. Furthermore, the guanidinated copolymers showed significantly decreased cytotoxicity, efficient trans fection at low charge ratios with pDNA, and good nuclear localization ability.
