By controlling the balance between salvage and catabolism of guanine in mammals, guanine deaminase(GDA) regulates both the guanine nucleotide supply and the production of xanthine, a substrate for the redoxsensitiveimmune regulator xanthine oxidase. GDA plays vital roles in purine homeostasis, neural developmentand immunity. Its expression is increased by oxidative stress and detection of GDA activity in serum orcerebrospinal fluid is regarded as a sensitive indicator of tissue trauma. This report describes a simple, versatile,robust and sensitive high-throughput GDA assay and some of its applications.
