HER Axis in Gastroesophageal Adenocarcinomas

The HER family of proteins consists primarily of four members. Human epidermal growth factor receptor-1 (HERD, also known as epidermal growth factor receptor (EGFR), is known to be overexpressed by a variety of epithelial malignancies including esophageal adenocarcinoma and to a lesser extent, gastric cancer. It is a transmembrane receptor with an external ligand binding domain and an internal ty-rosine kinase domain. Antibodies to the external domain are approved for treatment in colorectal cancer and head and neck cancers. Small molecule inhibitors of the tyrosine kinase are approved for use in lung cancer.
