Isolation, Purification and Identification of Genus Klebsiella and Framework of Systemic Tree

A pure strain was isolated from sludge of the wastewater treatment plant. It was Gram germ with 1.5-2 μm length. The strain was enriched, purified and separated gradually from liquid and solid cultivation under anaerobic conditions. The shape of colony and metabolic properties related to physiological and biochemical was studied on at the same time. It was identified according to its partial 16S rRNA gene sequence. Result showed it was genus klebsilla. This bacterium could degrade L-cysteine muriate and yield H2S. But it could not reduce the sulfate. So it was not SRB. The results showed that when separating and purifying SRB strain, disturance should be notedly caused by L-cysteine muriate or other organic matter with sulphur. It was necessary to further identify isolated strain by culture medium without organic sulfide to separate SRB from mixed samples.
