Large scale HLA-A, -B, and -DRB1 genotyping at intermediate level is performed using the rSSO method in our laboratory for donors who have joined the Canadian OneMatch Stem Cell and Marrow Network. High resolution HLA typing is performed on potentially matched donors using the Sequencing Based Typing Method (SBT). The laboratory frequently detects new alleles mainly by the SBT method. Here the identification of a novel HLA-A*01 null allele and a possible new HLA-Cw*0102 allele in two Caucasoid subjects which were miss assigned by SBT are described. HLA Intermediate Resolution typing was performed using the LABType rSSO A and C kits (One-Lambda Inc.). High Resolution typing was performed by the SBT method using Allele-SEQR HLA-A and HLA-C plus kits (Atria Genetics). HLA-A2 hemizygous sequencing was done using Allele-SEQR HLA-A2 group specific amplification (GSA). Sequencing was performed on an AB3130x1 Genetic Analyzer. The results were analyzed by the Assign-SBT software (Conexio Genomics).
