The present invention relates to a new analysis method of real-time PCR and kit capable of providing each combination of primers and probes in a one-to-one correspondence method that specifically amplifies and detects each allele of HLA-DRB1 and HLA-DRB3/4/5, allowing an easier and clearer analysis of the alleles using the results from a real-time PCR fluorescence curve, and of analyzing HLA-DRB1 and HLA-DRB3/4/5 alleles for a large amount of plasma more accurately and easily in a short time compared to conventional methods, allowing effective analysis on personal identification at forensic investigations and/or whether to transplant tissues during surgery at the genetic level. According to the present invention, more simultaneous, rapid and accurate detection of the alleles of HLA-DRB3/4/5 and HLA-DRB1 can be performed with high specificity and sensitivity from samples to be analyzed using real-time PCR, based on quantified data, thereby enhancing reliability and efficiency of the HLA-DRB1 and HLA-DRB3/4/5 genotypes when testing the individual identification at forensic investigations or whether to transplant tissues during surgery.
