Granulocytes are involved in recognizing and killing of fungi using Toll-like receptors (TLRs). We followed the effect of live, attenuated or antimycotic-treated conidia of A fumigatus and C. albicans on the expression of TLRs in granulocytes of children both at the mRNA and protein levels. Granulocytes were isolated from healthy adolescents using a standard Ficoll-paque protocol and incubated with A. fumigatus and C. albicans. The expression of TLR mRNA was checked using RTPCR, whereas the expression of TLR on the protein level was analyzed by using immunocytochemistry. A. fumigatus conidia up-regulated the expression of TLR9 by children granulocytes both at mRNA and protein levels. Caspofungin and amphotericin also stimulated the expression of TLR9, in contrast to the effect of C. albicans conidia alone, which down-regulated TLR9 expression. The present study demonstrated that TLR 9, play an important role in governing the function of granulocytes in fungal infections.
