Structure determination of RNA molecules at the atomic level is necessary to rationalize their multiple cellular functions. NMR spectroscopy is a powerful method to determine RNA solution structures up to 30kDa. Here I give an overview of the standard NMR techniques required for high-resolution structure determination of RNAs up to 15kDa. In addition, I will discuss which problems are encountered when studying larger RNAs by NMR (up to lOOkDa) and present a generally applicable approach for the high-resolution NMR structure determination of RNAs up to 30kDa.
