Cytokine Expression of Cultured Human Endothelial Cells Exposed to Biomaterials

In order to evaluate the roles vascular endothelial cells (EC) play in the thrombosis induced by biomaterials, the expression of mRNAs specific for two kinds of cytokine-interleukin-8 (IL-8) and tumor necrosis factor (TNF)-α were tested in vitro by cultured human umbilical vein endothelial cells (ECV304) exposed to different biomaterials, including PVC contained tin, polystyrene, polyurethane (PU50 & PU60), PTFE and ePTFE. The expressions of mRNAs were evaluated by RT-PCR with specific primers. The results showed (l)PU, PTFE and ePTFE, which were usually considered to have good hemocompatibility, however showed ability to induce the expression of cytokine in EC with different degrees; (2) In PVC contained tin, both IL-8 & TNF-α were expressed intensively, it demonstrated that it had a powerful effect on the immune and coagulative characteristics of EC with its poor hemocompatibility. The conclusion is the expression of cytokine IL-8 and TNF-α might indicate the hemocompatibility of biomaterials to some extent. There was also a good correlation between the expression of IL-8 & TNF-α(r=0.88571).
