To study the effects of vitamin C on the injury of alveolar macrophages expose to Ni_2O_3 in vitro and the expression of inducible nitric oxide synthase (iNOS). The alveolar macrophages were cultured with exposure to Ni_2O_3 in vitro. Meanwhile, vitamin C with different concentrations (25, 50 and 100μmol/L) were added in the medium, respectively. The detection of nitric oxide (NO), reactive oxygen species and activity of nitric oxide synthase (NOS) in alveolar macrophages were carried out. And the cell viability was determined after being exposed to Ni_2O_3 in vitro. The results revealed that vitamin c could decrease mortality and increase viability of alveolar macrophages. Active oxygen NO and NOS activity were decreased at the same time. Vitamin c at 50μmol/L and 100μmol/L had the most dramatics inhibitory effect. Vitamin c could antagonize the lipid peroxidation of alveolar macrophage induced by Ni_2O_3 which probably due to its antioxidant property.
