A stable transformation system was developed for the commercial apple cultivar 'New Jonagold' and the transgenic plants carrying antisense gene of ACC oxidase were obtained. Leaf strips excised from in vitro grown plants were co-cultured for 3 d with Agrobacterium tumefaciens strain EHA105. The cocultivated leaf strips were incubated on MS medium supplemented with BA 6.0mg/l + IAA 0.8mg/l + cefotaxime 200mg/l for 2 d without selection, and then on MS medium containing TDZ 3.0mg/l + IAA 0.5mg/l + cefotaxime 200mg/l + kanamycin 40mg/l for selection. The transformed buds appeared on the explants obviously after the leaf strips incubated on selectable medium for 3 weeks. Regeneration percentage of transformation buds averaged 1.2%. Transformation was confirmed by PCR analysis. The transgenic plants were transplanted successfully and grow well.
