We proposed a novel method to synthesize biologically active peptides utilizing an extractive hydrolysis of proteins in an aqueous two-phase system, and applied it to the thermolysin-catalyzed hydrolysis of zein (com protein, Zea mays L.) to synthesize angiotensin converting enzyme (ACE) inhibitory peptides. Zein and its enzymatic hydrolysates (angiotensin converting enzyme inhibitory peptides) distributed to poly(ethyleneglycol) (PEG) and dextran (DEX) rich phases, respectively, in an aqueous PEG/DEX two-phase system. The ACE inhibitory peptides are selectively distributed in a DEX rich phase.
