Tumor metastasis is a complicated process and therefore it is not surprising that multiple proteins have been proposed to be involved. This was often based on correlations between metastatic capacity and expression levels of the protein, but involvement was usually difficult to prove. For a set of model lymphoma cell lines, we have assessed the role in metastasis of several cell surface as well as signaling molecules using different molecular biological approaches. In this chapter, the results are discussed briefly, but the emphasis is on the advantages and disadvantages of the different approaches and on potential pitfalls. The migration mechanisms of T-lymphoma cells as well as normal T lymphocytes, deduced from these experiments, will be discussed in a separate chapter.
