Although genetic alterations have been implicated in the process of human gastric carcinogenesis, the principle carcinogenic mechanisms are not fully understood. This study was conducted a proteomic approach to search for genes involved in gastric carcinogenesis. They might serve as diagnostic markers. We identified 9 proteins with increased expression and 14 proteins with decreased expression in gastric carcinomas, suggesting links between dysfunctions of the proteins and gastric carcinogenesis. Importantly, immunohistochemical analyses were consistent with proteomic data, which confirmed that the levels of expression of MAD1L1, HSP27, and CYR61 were altered in gastric carcinoma tissues. Therefore, our study suggested that (i) the proteins identified in this study could be useful diagnostic markers and that (ii) a proteomics-based approach is useful for developing a more complete picture of the pathogenesis and function of human gastric carcinomas.
