Inducing of Human IgE Antibodies by In Vitro Immunization

To induce antibodies in vitro we used a medium containing enriched RDF supplemented with human plasma, fetal calf serum, IL-2, IL-4, IL-6, and muramyl dipeptide. Human lymphocytes were cultivated at a final density of 1 x 106 cells/ml in the medium of 96- or 24-well plates in a CO_2 incubator at 37癈 for 10 days. After 10 days, about 100 ng/ml of IgE antibody was secreted into the medium. The technique requires the use of plasma and lymphocytes obtained from the same donor. This IgE-inducing system was applied while surveying a variety of food ingredients that alleviate pollenosis. Oolong tea and honey had the highest alleviating activity among the 190 food ingredients tested. This IgE-inducing system is expected to be extremely useful in studying allergy mechanisms and surveying allergy-alleviating products. This system can be also applied for establishing cell lines secreting antigen-specific IgE antibodies by immortalizing B cells after antibody induction.
