The provision of lipid components in aqueous based tissue culture media is difficult due to their limited solubility. Serum supplementation circumvents this but introduces contamination and consistency problems and non-serum lipid supplements are poor substitutes. To overcome these issues we have developed a new biomimetic lipid supplement, synthetic low density lipoprotein (sLDL), which is prepared by a cGMP compliant microfluidisation process using animal free components. sLDL is physically and chemically stable, filterable and can be prepared to contain a range of triglyceride and cholesterol ester components. sLDL cellular uptake is concentration, time and temperature dependent and undergoes competition with native LDL indicating a receptor dependent uptake via the LDL receptor. The cellular uptake permits sLDL to act as a lipid supplement and support the proliferation of cells in serum free media systems. sLDL therefore represents a novel biomimetic lipid supplement for serum free tissue culture, the exploration of cellular dietary lipid preferences and cellular delivery.
