The new MS-based strategy for analysis of sulfated GAGs is facile, specific (with distinct chlorine-isotope trait markers), sensitive (with a detection limit of ~70 pg), and potentially useful for rapid and high-throughput diagnosis of MPS. Sulfated oligosaccharides were selectively extracted using polyarginine-coated nanodiamonds (PA-coated NDs), degraded to disaccharides through microwave-assisted depolymerization, mixed with deuterium-labeled internal standards for quantitative analysis by MALDI-TOF MS using the N-(1-naphthyl) ethylenediamine dihydrochloride (NEDC) matrix.
